Wilkin Ramírez was signed by the Detroit Tigers on February 5, 2003 by scout Ramon Pena. He made his pro debut that summer with the GCL Tigers as a third baseman. He was second in the Gulf Coast League with 7 triples and fourth with 35 RBIs, while pacing third baseman with 86 assists and 21 errors. However, he missed the entire next season following shoulder surgery. In 2005, with the West Michigan Whitecaps, he earned a spot on the Midwest League All-Star team after hitting .262 with 16 home runs, despite leading the league with 143 strikeouts. However, the following summer, he only played in 66 games for the Lakeland Tigers after suffering a shin contusion.

Ramírez began 2007 back with Lakeland and was moved from third to the outfield. He stole 28 bases with the club before being promoted to the AA Erie SeaWolves in late July. He spent most of the following year with Erie and was named an Eastern League All-Star and the team's Player of the Year after hitting .303 with 7 triples and 26 stolen bases. He also briefly played at the AAA level in June, making 11 appearances for the Toledo Mud Hens. He began 2009 back with the Mud Hens and was briefly called up by the Tigers in May while Magglio Ordonez was on the family medical emergency list. He appeared in one big league game, earning a start as designated hitter, and hit a home run, becoming the first Tiger to homer in his debut in more than thirty years. When Ordonez returned, he was sent back down to the Mud Hens.
