The son of big leaguer Lee Mazzilli, infielder L.J. Mazzilli was selected by the Minnesota Twins in the ninth round of the 2012 amateur draft but did not sign, opting to return to the University of Connecticut instead. He was then chosen by the New York Mets in the fourth round of the 2013 amateur draft. He soon signed and made his pro debut for the Brooklyn Cyclones, going 2-for-4 against the Staten Island Yankees on June 17th. he hit .278/.329/.381 in 70 games that first season as Brooklyn's starting second baseman.

In 2014, Mazzilli played for three different teams, starting the year with the Savannah Sand Gnats of the South Atlantic League, for whom he hit .292 in 66 games. That earned him a mid-season promotion to the St. Lucie Mets of the Florida State League, where he hit .312 in 64 games. the Mets rewarded him for his excellent season by giving him an opportunity to help out the AAA Las Vegas 51s for the playoffs. He got into one regular-season game for them, going 1 for 5. Altogether, his batting line was .301/.361/.440 in 131 games, with 29 doubles, 11 homers and 79 RBIs and as many runs scored. However, just when it seemed Mazzilli was on a fast track to the big leagues, his career hit a bump that off-season when he was handed a 50-game suspension for a second positive test for an unspecified drug of abuse.
