The 2005 Major League Baseball Amateur Draft was held on June 7 and June 8, 2005. The first overall pick was Justin Upton by the Arizona Diamondbacks.

First pick Upton is the brother of of B.J. Upton who had been selected with the second overall pick in 2002 by the Tampa Bay Devil Rays. Golden Spikes Award winner Alex Gordon of the University of Nebraska was drafted with the number two selection by the Kansas City Royals. Wade Townsend, who had been drafted the previous year by the Baltimore Orioles but could not reach a contract agreement, was drafted by Tampa Bay.

Collegiate players accounted for two-thirds of the first round selections and all but the first and tenth of the top ten picks. 13 pitchers were selected in the first round.

Joey Devine became the first player from the draft to play in the MLB when he debuted on August 20, 2005.
